DIVISION of BIOLOGICAL SCIENCES PROTEIN SEQUENCING FACILITY

 

The Division of Biological Sciences Protein Sequencing Facility is located in Room 2121 in Bonner Hall on the Revelle Campus of the University of California at San Diego in La Jolla, California (See map). The Facility operates an Applied Biosystems PROCISE 494HT Protein Sequenator.   • Description of Services:
    • Provide N-terminal sequencing of protein and peptide samples
      • We accept samples in solution, attached to beads, or transferred from polyacrylamide gels to PVDF membranes.
      • Typical requirement is 5 pmol of purified material, although the absolute sensitivity of the instrument is on the order of 0.5 pmol. We will gladly attempt lower amounts, if you are unable to obtain larger quantities
    • Samples in solution are usually adsorbed to PVDF membranes using an Applied Biosystems PROSORB device
    • Each sample typically take 3-6 business days to process; you will receive results as a pdf file by email with sequence and chromatograms
  • Charges for Services/Payment Information:
    • University of California Users:
      • $31 start up fee (effective 3/16/09) for each sample.
      • $31/cycle Amino Acid Residue for amino acids 1-10 (minimum charge is 5 residues) for each sample.
      • After 10 amino acids, residues 11 and beyond are charged half-price for each sample.
    • For Users Outside UC System working at a Non-Profit Organization:
      • $31 start up fee (effective 3/16/09) for each sample.
      • $35.50/Amino Acid Residue for amino acids 1-10 (minimum charge is 5 residues) for each sample.
      • After 10 amino acids, residues 11 and beyond are charged half-price for each sample.
    • For Users Outside UC System working at a 'For-Profit' Organization:
      • $45 start up fee (effective 3/16/09) for each sample.
      • $46/Amino Acid Residue for amino acids 1-10 (minimum charge is 5 residues) for each sample.
      • After 10 amino acids, residues 11 and beyond are charged half-price for each sample.
    • How to Pay:
      • UC users can pay using index numbers/purchasing accounts, or purchase orders. Users outside the UC system can only pay using a purchase order.
      • If you are using a purchase order, we will need a hardcopy of it. You can xerox a copy and send it with your sample, fax it to 858-534-1492, or scan and email it to proteinsequencer@gmail.com.
  • Requirements for Samples:
    • Approximately 85% purity of sample
    • Sample can be in solution, attached to a bead, or transferred from gel to PVDF membrane and stained with Coomassie blue, Ponseau S, or Amido black (procedures for pvdf transfer can be found online). Please do not use nitrocellulose as it is soluble in sequencing reagents.
    • Staining is not absolutely necessary but will enable us to ensure the purity of your sample.
  • How to Submit Your Sample for Sequencing:
    • Download Sample Submission Form, fill it out and send or bring with your sample(s).
    • Bring your sample, sample submission form, and hardcopy of the purchase order (if applicable, not needed if you are UC user) to Bonner 2121 in person Mon-Fri 8 am-5pm OR mail using Federal Express to the following address: Protein Sequencer Facility,
